Our experimental data confirmed that S. shinii and S. succinus exhibited significant acaropathogenic properties against R. microplus, as demonstrated by the tracking of fluorescent-labeled bacteria within the engorged-tick body. Our experiments revealed that both bacterial species could infect the hemolymph, salivary glands, and vestibular vagina of the tick, inducing histological changes in the affected organs that may impair feeding as well as reproductive capabilities. Gené's organ infection was detected only in S. succinus. Biofilm eradication tests demonstrated a significant combined beneficial effect (Bliss independence model, CI < 1) as phages improved carrier bacteria colonization by 1.2-fold and carrier bacteria facilitated phage infection by 1.4-fold. Transcriptomic analysis showed that phoresy significantly enhanced motility (e.g., fliC and pilD genes) and energy metabolism (e.g., pgm and pgk genes) of carrier bacteria and suppressed the defense system (e.g., MSH3 and FLS2 genes) and energy metabolism (e.g., petB and petC genes) of pathogens. Metabolomics analysis revealed that the phoresy system stimulated the secretion of beneficial metabolites (e.g., flavonoid and tropane alkaloid) that could enhance stress response and phyllosphere protection in soybeans. Our analysis identified 108 Cecropin genes from 105 insect transcriptomes, covering all major hexapod lineages. We validated the gene sequences and synthesized mature peptides for three identified Cecropin genes. Through minimal inhibition concentration and agar diffusion assays, we confirmed that these peptides exhibited antimicrobial activities against Gram-negative bacteria. Similar to the known Cecropin, the three Cecropins adopted an alpha-helical conformation in membrane-like environments, efficiently disrupting bacterial membranes through permeabilization. Importantly, none of the three Cecropins demonstrated cytotoxicity in erythrocyte hemolysis tests, suggesting their safety in real-world applications. We show explicitly that these strains demonstrate high acidification rate, completely repress the growth of E. coli, S. aureus and to a lesser extent P. aeruginosa as well as exhibit appropriate probiotic and milk-fermenting properties. Moreover, in contrast to the approved probiotic strain Lactobacillus plantarum 8PA3, the new isolates were able to efficiently eradicate preformed biofilms of these pathogens and prevent bacterial spreading originating from the biofilm. We suggest these strains as potential additives to the pre-cultures of conventional LAB strains as efficient tools targeting foodborne pathogens in order to prevent food contamination from either seeded raw material or biofilm-fouled equipment. Then, a database of the intestinal transcriptome of the host fruit fly was established and analyzed using the Illumina HiSeq Platform. The gut bacteria shifted from Gram negative to Gram positive after antibiotic feeding. Antibiotics lead to a reduction in gut bacteria, particularly Gram-positive bacteria, which ultimately reduced the reproduction of the host flies. Ten immunity-related genes that were differentially expressed in the response to intestinal bacterial community changes were selected for qRT-PCR validation. Peptidoglycan-recognition protein SC2 gene (PGRP-SC2) was one of the 10 immunity-related genes analyzed.
